It is water repellent, not water "proof." It should be fine in rain but not submerged.

When this is empty, can it be compressed or folded for smaller profile, making it less bulky while riding?

There are straps on each side of the backpack that you can tight to flatten the backpack. You won’t be able to roll it up but you can make the backpack flat.

Does the fender have to be removed to ensure the Onewheel fits? Or can it stay on?
