Will this work with networking cables (5e) run through the walls to wall plates with an ethernet cable from the wall plate to the adapter?

The answer by Wes is incorrect. He must have some other problem. We use these in our church to transmit HDMI over 100' CAT5e cables, and they work perfectly.

Do these come as pairs?

YES, they come in specificlly matched pairs, so when you replace these, you need to replace BOTH the sender unit and the receiver unit. Even if only of your original Extenders has blown out, you need to replace both because they are matched to talk to each other. Good luck.
