Q: I have to join tubing that is 5/16 id, 9/16 od that will carry co2 gas from a tank to a keg coupler. what size would be best?

Mostly depends on how soft the hose is and the pressure. Soft 5/16 hose will easily push onto a 3/8 barb.

Q: Will this work with 3/4 polybutylene irrigation line?

No. These have an external dimension of 3/8. Measure the internal diameter of your tube and select a connector with that measurement.

Q: Is this toxic? I want to use it for my fridge water

It's not toxic, has the NSF/ANSI 61 standard. Safe for potable water.
