Q: Does this work in a cr-10 v2?

It worked on my CR10SPRO V2. NOT SURE IF ITS THE SAME OR NOT.

Q: Will this work on a creality ender 3 s1 pro sprite extruder?

This is for the CR-10S Pro, CR-10S Pro V2, and the CR-10 Max printers. There are unofficial ways to make this work with a Sprite extruder added to one of these 3 printers, but this is the OEM equivalent of the breakout board used for the ribbon cable from the motherboard up to the X axis which then uses smaller separate connectors for extruder/hotend components.
