Q: Which is more effective; wet or dry brushing; feels more sensitive when wet brush?

Dry brushing gives better exfoliation. Wet brushing works if you have sensitive skin that's irritated if you dry brush

Q: Will the handle turn black after using it for a while?

Thus far, mine hasn't developed mold or mildew anywhere on it. I shake it off, hang it on a hook in the shower, and leave the sliding shower doors open so there's ventilation.

Q: What is the best way to store it and keep it clean/prevent mold when not in the shower?

Mold growth is a function of poor air circulation preventing the removal of water from the air and surfaces in the bath. Hang the brush somewhere it can dry.
