It senses moisture through a painted surface.

Q: Can this be used on concrete and drywall?

Yes, the device has four modes. Wall, masonry, soft wood, hard wood. Wall is drywall.

Q: Can this be used to locate a leak behind drywall. The drywall is not showing wetness so the water is probably running down a stud or outside wall.

This tool will measure the moisture content of any building material including wood and drywall. If the test indicates a high moisture content in just one spot, you could probably conclude that there was a leak at that point.

Q: Will this unit work on a plywood sub floor, and hardwood flooring to verify the hardwood has properly aclimated for installation?

In order to get accurate readings you will need to make sure the two prongs on the top make contact with the area you wish to measure for moisture. The two prongs are pretty small and the tips are sharp. I was able to obtain readings on wet drywall, plywood and ceramic floor tile. As long as contact was made with the points it worked for me and the price is reasonable.
